Leflunomide is a disease-modifying antirheumatic drug (DMARD) used to treat rheumatoid arthritis. It works by inhibiting the enzyme dihydroorotate dehydrogenase (DHODH), which is involved in the synthesis of pyrimidines, essential building blocks for DNA and RNA. This inhibition slows the proliferation of lymphocytes, key cells in the immune system that drive inflammation in rheumatoid arthritis.
