Brand: Soriatane
Acitretin is a retinoid, a synthetic derivative of vitamin A, primarily used for the treatment of severe psoriasis. It works by normalizing the growth and differentiation of skin cells. Acitretin belongs to the class of systemic retinoids and acts by binding to retinoid receptors in the skin, influencing gene expression and reducing inflammation and abnormal cell proliferation.
For severe psoriasis resistant to other treatments.
Acitretin can cause severe birth defects. Women of childbearing potential must not take acitretin if pregnant or planning to become pregnant. Strict contraception is required during treatment and for at least 3 years after discontinuation due to the long elimination half-life of acitretin and its active metabolite.
Outcome:
Increased formation of etretinate, a long-acting metabolite with teratogenic potential.
Mechanism:
Alcohol can interfere with the metabolism of acitretin.
Outcome:
Altered metabolism of both drugs
Mechanism:
Enzyme induction/inhibition
Outcome:
May reduce absorption of acitretin
Mechanism:
Changes in gastric pH
Most likely new formulation: Topical acitretin for localized psoriasis treatment (2025, 30% confidence)
Based on current usage trends and clinical trial data, there is a low (5%) likelihood of any major regulatory changes for acitretin in the next 2 years.
