Aminoglutethimide is an inhibitor of steroid synthesis, primarily used in the treatment of Cushing's syndrome and breast cancer. It works by blocking the conversion of cholesterol to pregnenolone, thereby reducing the production of adrenal corticosteroids and androgens. While effective, it has been largely replaced by newer, more selective agents due to its potential for significant side effects.

Increased risk of CNS depression
Mechanism:
Additive CNS depressant effects.
Outcome:
Potential for gastrointestinal irritation
Mechanism:
Additive gastrointestinal effects.
Most likely new formulation: extended-release formulation (Year: Unlikely, 1% confidence)
Based on historical usage patterns and the availability of newer agents, the likelihood of new aminoglutethimide formulations being developed is less than 5%.
