Bleomycin is a glycopeptide antibiotic with antineoplastic activity. It is used to treat various types of cancer by causing DNA strand breaks, leading to cell cycle arrest and apoptosis. It belongs to the antitumor antibiotic class and is cell cycle-specific, primarily affecting the G2 and M phases.
For treatment of various cancers including Hodgkin's lymphoma, non-Hodgkin's lymphoma, testicular cancer, squamous cell carcinoma, and malignant pleural effusion.
Pulmonary toxicity, including pneumonitis and pulmonary fibrosis, can occur and may be fatal. Patients with underlying pulmonary disease, prior radiation therapy to the chest, or receiving high cumulative doses are at increased risk. Monitor closely for pulmonary symptoms and perform pulmonary function tests.
Outcome:
Increased risk of pulmonary toxicity
Mechanism:
Synergistic lung damage
Outcome:
Reduced vaccine efficacy
Mechanism:
Immunosuppression
Outcome:
Delayed absorption

Most likely new formulation: Liposomal bleomycin for reduced pulmonary toxicity (2025, 30% confidence)
Based on historical usage patterns and current research trends, there is a low ( <5%) likelihood of significant regulatory changes for bleomycin in the next 5 years.
