Brand: Clolar
Clofarabine is a purine nucleoside antimetabolite used to treat certain types of leukemia. It belongs to a class of drugs known as antineoplastics. Clofarabine works by interfering with DNA synthesis and repair, ultimately leading to cell death in rapidly dividing cancer cells.
Used to treat relapsed or refractory acute lymphoblastic leukemia in children.
Severe and sometimes fatal hepatotoxicity, renal failure, and capillary leak syndrome have been reported. Closely monitor patients for these adverse reactions.
Outcome:
Increased risk of myelosuppression
Mechanism:
Additive myelosuppressive effects
Outcome:
Increased risk of clofarabine toxicity
Mechanism:
Inhibition of clofarabine metabolism
Outcome:
Decreased clofarabine absorption
Mechanism:
Chelation or altered pH
Most likely new formulation: Liposomal clofarabine for improved delivery and reduced toxicity (Year: 2026, 60% confidence)
Based on current usage and clinical trial data, there is a 70% likelihood of clofarabine remaining a key treatment option for relapsed or refractory acute lymphoblastic leukemia.
