Brand: Dantrium, Ryanodex, Revonto
Dantrolene is a postsynaptic muscle relaxant that directly interferes with calcium release from the sarcoplasmic reticulum of skeletal muscle. It is primarily used to treat malignant hyperthermia (MH), a rare but life-threatening condition triggered by certain anesthetics. Dantrolene is also used to manage chronic spasticity disorders such as multiple sclerosis, cerebral palsy, and spinal cord injury.
For treating malignant hyperthermia and managing chronic spasticity.
Hepatotoxicity: Dantrolene can cause serious and potentially fatal liver damage. Liver function tests should be performed before and during treatment.
Outcome:
Hyperkalemia, cardiac arrest
Mechanism:
Additive effects on calcium channels
Outcome:
Increased risk of hyperkalemia
Mechanism:
Additive effects on potassium homeostasis
Outcome:
Reduced dantrolene absorption
Mechanism:
Chelation with divalent cations
Most likely new formulation: Extended-release oral formulation (2025, 80% confidence)
Based on current usage trends and emerging clinical data, there is a 70% likelihood of expanded indications for dantrolene in the treatment of other muscle disorders within the next 5 years.
