Entecavir is a nucleoside analogue reverse transcriptase inhibitor (NRTI) used in the treatment of chronic hepatitis B virus (HBV) infection. It works by interfering with the virus's ability to replicate its DNA, thus reducing the viral load and slowing the progression of liver disease. Entecavir is generally well-tolerated but can have rare serious side effects such as lactic acidosis and severe liver problems.
For the treatment of chronic hepatitis B infection.
Outcome:
Increased risk of lactic acidosis and liver toxicity
Mechanism:
Both drugs are NRTIs and can have additive mitochondrial toxicity.
Outcome:
Increased risk of nephrotoxicity
Mechanism:
Additive renal toxicity.
Outcome:
Reduced entecavir absorption
Mechanism:
Binding of entecavir in the gut.
Most likely new formulation: Combination therapy with other antivirals (2025, 75% confidence)
Based on current usage trends and clinical trial data, there is a 90% likelihood that entecavir will maintain its approved status for chronic hepatitis B treatment.
