Estradiol is a naturally occurring estrogen, the primary female sex hormone. It is crucial for developing and maintaining the female reproductive system and secondary sexual characteristics. Estradiol also plays a role in bone health, cholesterol metabolism, and cognitive function. Synthetic forms of estradiol are available as pharmaceuticals and used in hormone replacement therapy (HRT) for menopause symptoms, female hypogonadism, and certain types of breast and prostate cancer.

Estrogens increase the risk of endometrial cancer in postmenopausal women. The addition of a progestin reduces this risk. Estrogens, with or without progestins, should be used at the lowest effective dose and for the shortest duration consistent with treatment goals and risks for the individual woman.
