Leflunomide is a disease-modifying antirheumatic drug (DMARD) used to treat rheumatoid arthritis. It works by inhibiting the enzyme dihydroorotate dehydrogenase (DHODH), which is involved in pyrimidine synthesis, a pathway crucial for the rapid proliferation of lymphocytes (cells involved in immune responses). By suppressing this pathway, leflunomide reduces inflammation and slows joint damage in rheumatoid arthritis.
