LY146032, better known as rosuvastatin calcium, is a synthetic statin used primarily to lower cholesterol levels in the blood. It belongs to the class of HMG-CoA reductase inhibitors, which work by blocking the enzyme responsible for producing cholesterol in the liver. This action reduces the amount of LDL cholesterol ('bad' cholesterol) and triglycerides, while slightly increasing HDL cholesterol ('good' cholesterol). Rosuvastatin is prescribed to reduce the risk of cardiovascular events like heart attack and stroke, especially in individuals with high cholesterol, diabetes, or a family history of heart disease.
Lowers cholesterol to reduce the risk of cardiovascular disease.
Outcome:
Increased rosuvastatin levels, increased risk of myopathy
Mechanism:
Inhibition of CYP3A4 metabolism
Outcome:
Potential for altered INR
Mechanism:
Possible interaction with vitamin K metabolism
Outcome:
Slight decrease in rosuvastatin absorption
Mechanism:
Binding to rosuvastatin
Most likely new formulation: sustained-release formulation (Year 2026, 70% confidence)
Based on current usage trends and clinical trial data, there is a low probability ( <5%) of any major regulatory changes to rosuvastatin's labeling in the next 2 years.
