Nefazodone is an atypical antidepressant that was primarily used to treat major depressive disorder (MDD). It belongs to the phenylpiperazine class of antidepressants and works by inhibiting the reuptake of serotonin and norepinephrine, two neurotransmitters believed to play a role in mood regulation. Nefazodone also blocks certain serotonin receptors, which may contribute to its unique side effect profile compared to other antidepressants.

Severe liver damage, including liver failure requiring transplantation or leading to death, has been reported with nefazodone. Nefazodone therapy should be discontinued in patients who develop jaundice or laboratory evidence of liver dysfunction.
