Nitazoxanide is a synthetic nitrothiazolyl-salicylamide derivative with broad-spectrum antiparasitic and antiviral activity. It is used to treat various protozoal and helminthic infections, including giardiasis and cryptosporidiosis. Its mechanism of action involves interference with pyruvate:ferredoxin oxidoreductase (PFOR) enzyme-dependent electron transfer reactions, which are essential for anaerobic energy metabolism in these parasites. Nitazoxanide also exhibits antiviral activity against a range of RNA and DNA viruses, including influenza, rotavirus, and norovirus, potentially through inhibition of viral RNA-dependent RNA polymerase.
