Brand: Eloxatin
Oxaliplatin is a platinum-based antineoplastic agent used in the treatment of colorectal cancer. It belongs to a class of drugs known as alkylating agents, specifically platinum analogues. Oxaliplatin exerts its cytotoxic effects by forming intra- and inter-strand crosslinks in DNA, which inhibits DNA replication and transcription, ultimately leading to cell death. It is often used in combination with other chemotherapeutic agents, such as 5-fluorouracil and leucovorin (FOLFOX regimen) or capecitabine (XELOX regimen).
Used to treat advanced colorectal cancer, often in combination with other chemotherapy drugs.
Acute hypersensitivity reactions, including anaphylaxis, can occur. Severe and sometimes fatal hepatic sinusoidal obstruction syndrome (SOS) has been reported. Peripheral sensory neuropathy can occur and may be cumulative. Myelosuppression may occur.
Outcome:
Increased risk of myelosuppression
Mechanism:
Additive myelosuppressive effects
Outcome:
Increased risk of ototoxicity and nephrotoxicity
Mechanism:
Additive toxicity
Outcome:
May slightly reduce absorption
Mechanism:
Altered pH
Most likely new formulation: Liposomal oxaliplatin for improved efficacy and reduced toxicity (Year 2026, 60% confidence)
Based on ongoing clinical trials and real-world data analysis, there is a 70% likelihood of expanded indications for oxaliplatin in other solid tumors within the next 5 years.
