Brand: Vioxx
Rofecoxib, formerly marketed under the brand name Vioxx, is a nonsteroidal anti-inflammatory drug (NSAID) of the coxib class that was used to treat pain, inflammation, and fever. It selectively inhibits cyclooxygenase-2 (COX-2), an enzyme involved in prostaglandin synthesis. While effective for its intended uses, rofecoxib was withdrawn from the market in 2004 due to increased cardiovascular risks.
Used to treat osteoarthritis, rheumatoid arthritis, acute pain, and menstrual pain.
Outcome:
Increased bleeding risk
Mechanism:
Rofecoxib can displace warfarin from plasma proteins, increasing its free concentration.
Outcome:
Increased methotrexate toxicity
Mechanism:
Rofecoxib can reduce renal clearance of methotrexate.
Outcome:
Reduced absorption of rofecoxib
Mechanism:
Antacids can alter the pH of the stomach, affecting rofecoxib absorption.
No new formulations of rofecoxib are expected.
Due to its withdrawal, there is a 0% likelihood of rofecoxib being reintroduced to the market.
