Sulfonamides are a class of synthetic antimicrobial agents containing the sulfonamide group. They were among the first antibiotics discovered and used widely in the mid-20th century. While largely replaced by newer antibiotics for many infections, they remain valuable for treating specific conditions, notably urinary tract infections. Sulfonamides act by inhibiting bacterial synthesis of dihydrofolic acid, essential for bacterial growth.
Used primarily to treat urinary tract infections and certain other bacterial infections.
Outcome:
Increased risk of bleeding
Mechanism:
Sulfonamides can displace warfarin from plasma proteins, increasing its free concentration.
Outcome:
Hyperkalemia
Mechanism:
Sulfonamides can reduce renal potassium excretion.
Outcome:
Reduced absorption of sulfonamides
Mechanism:
Antacids can increase the pH of the stomach, reducing sulfonamide absorption.
Most likely new formulation: Extended-release combination with a beta-lactamase inhibitor (2025, 60% confidence)
Based on current usage trends and emerging resistance patterns, there is a 20% likelihood of increased restrictions on sulfonamide use for uncomplicated UTIs within the next 5 years.
