Tenofovir is an antiviral medication used to treat chronic hepatitis B and prevent and treat HIV/AIDS. It belongs to a class of drugs called nucleotide reverse transcriptase inhibitors (NRTIs) or, more specifically when formulated with other antivirals, nucleoside/nucleotide reverse transcriptase inhibitor backbone. Tenofovir works by blocking the reverse transcriptase enzyme, which HIV and hepatitis B virus need to replicate their genetic material. It is available in two main forms: tenofovir disoproxil fumarate (TDF) and tenofovir alafenamide fumarate (TAF). TAF is generally preferred due to its improved safety profile, particularly regarding kidney and bone health.
Used to treat chronic hepatitis B and prevent and treat HIV/AIDS.
Outcome:
Increased didanosine levels, leading to increased risk of didanosine-related adverse events.
Mechanism:
Tenofovir can decrease the renal clearance of didanosine.
Outcome:
Increased tenofovir levels
Mechanism:
Ritonavir inhibits the metabolism of tenofovir.
Outcome:
Increased tenofovir absorption
Mechanism:
Enhanced bioavailability
Most likely new formulation: Long-acting injectable Tenofovir (2025, 75% confidence)
Based on current usage patterns and clinical trial results, there is a 90% likelihood that Tenofovir will remain a first-line treatment for HIV and Hepatitis B in the next 5 years.
