Tenoxicam is a non-steroidal anti-inflammatory drug (NSAID) belonging to the oxicam class. It works by inhibiting cyclooxygenase (COX) enzymes, which are responsible for producing prostaglandins, substances that mediate pain, inflammation, and fever. Tenoxicam's long half-life allows for once-daily dosing.
For the relief of pain and inflammation associated with osteoarthritis, rheumatoid arthritis, and other musculoskeletal disorders.
Tenoxicam, like other NSAIDs, may increase the risk of serious cardiovascular thrombotic events, myocardial infarction, and stroke, which can be fatal. This risk may increase with duration of use. Patients with cardiovascular disease or risk factors for cardiovascular disease may be at greater risk. Tenoxicam is contraindicated for the treatment of peri-operative pain in the setting of coronary artery bypass graft (CABG) surgery. NSAIDs can also cause an increased risk of serious gastrointestinal adverse events, including bleeding, ulceration, and perforation of the stomach or intestines, which can be fatal. These events can occur at any time during use and without warning symptoms. Elderly patients are at greater risk for serious gastrointestinal events.
Outcome:
Increased risk of bleeding
Mechanism:
Tenoxicam can displace warfarin from protein binding sites, increasing its free concentration.
Outcome:
Increased risk of gastrointestinal bleeding
Mechanism:
Synergistic effect on gastric mucosa irritation.
Outcome:
Reduced tenoxicam absorption
Mechanism:
Altered pH in the stomach.
Most likely new formulation: Topical cream for localized pain relief (Year 2025, 70% confidence).
Based on current usage trends and safety profile, tenoxicam is expected to maintain its current regulatory status with a 90% likelihood.
